MTT assay is a colorimetric assay for assessing cell metabolic activity. NAD(P)H-dependent cellular oxidoreductase enzymes may, under defined conditions, reflect the number of viable cells present. These enzymes are capable of reducing the tetrazolium dye MTT 3-(4,5-dimethylthiazol-2-yl)-2,5-phenyltetrazolium bromide to its
insoluble formazan, which has a purple color.
Tetrazolium dye assays can also be used to measure cytotoxicity or cytostatic activity of potential medicinal agents and toxic materials. MTT assays are usually done in the dark since the MTT reagent is sensitive to light.

Preparation:
Solubility: in water (10 mg/ml), ethanol (20 mg/ml) and 2-methoxyethanol (20mg/ml).
It is also soluble in buffered salt solutions and culture media (5 mg/ml).
Storage/Stability:
MTT is light sensitive. Reconstituted MTT solution is stable for at least 6 months when stored frozen (-20 °C).
Storage of a reconstituted MTT solution at 2 - 8 °C for more than 2 weeks may cause decomposition and yield erroneous results.
